چکیده
Regulatory T cells (Treg) restrain skin inflammation under healthy conditions and after immune challenges. Adult human contains 20 billion 1-3 are highly suppressive FOXP3+ Treg. We observed inflammatory eruptions in CTCL patients treated with alemtuzumab (αCD52), an antibody that purges recirculating from skin. αCD52-induced eczematous dermatitis had 5-fold lower Treg numbers (p<0.0001) 7-fold frequencies compared to age matched controls dermatitis, suggesting αCD52 may be inducing by depleting Immunostaining of showed only 18% expressed resident memory cell (TRM) markers (CD69 and/or CD103) 70% FOXP3- cells. Flow cytometry studies confirmed a second method 16% TRM markers, 53% 72% conventional CD4 CD8 cells, respectively. Only 2% total were Treg; 98% Single RNA sequencing enriched for recirculation associated genes. To functionally study the Treg, we biopsied before treatment αCD52. In all cases, reduced both absolute number relative (89% depleted), confirming preferentially recirculating. summary, report resident. The spontaneous reactions occur αCD52, medication depletes as they enter bloodstream, demonstrates presence is required suppression inflammation.

Activation-induced FOXP3 in human T effector cells does not suppress proliferation or cytokine production.
Forkhead box P3 (FOXP3) is currently thought to be the most specific marker for naturally occurring CD4(+)CD25(+) T regulatory cells (nTregs). In mice, expression of FoxP3 is strictly correlated with regulatory activity, whereas increasing evidence suggests that in humans, activated T effector cells (Teffs) may also express FOXP3. In order to better define the role of FOXP3 in human Teff cells,...
متن کاملFunctionally distinct subsets of human FOXP3+ Treg cells that phenotypically mirror effector Th cells.
FOXP3+ regulatory T (Treg) cells are a broadly acting and potent anti-inflammatory population of CD4+ T cells essential for maintaining immune homeostasis and preventing debilitating autoimmunity. Based on chemokine receptor expression, we identified distinct populations of Treg cells in human blood expected to colocalize with different Th cell subsets. Although each population was functionally...
متن کاملIncreased CD4+Foxp3+ T cells in BAFF-transgenic mice suppress T cell effector responses.
The cytokine B cell activation factor of the TNF family (BAFF) is considered to perform a proinflammatory function. This paradigm is particularly true for B cell-dependent immune responses; however the exact role for BAFF in regulating T cell immunity is ill-defined. To directly assess the effect of BAFF upon T cells, we analyzed T cell-dependent immune responses in BAFF-transgenic (Tg) mice. W...
متن کاملResident CD141 (BDCA3)+ dendritic cells in human skin produce IL-10 and induce regulatory T cells that suppress skin inflammation
Human skin immune homeostasis, and its regulation by specialized subsets of tissue-residing immune sentinels, is poorly understood. In this study, we identify an immunoregulatory tissue-resident dendritic cell (DC) in the dermis of human skin that is characterized by surface expression of CD141, CD14, and constitutive IL-10 secretion (CD141(+) DDCs). CD141(+) DDCs possess lymph node migratory c...
متن کاملDifferential Responses of Human Regulatory T Cells (Treg) and Effector T Cells to Rapamycin
BACKGROUND The immunosuppressive drug rapamycin (RAPA) promotes the expansion of CD4(+) CD25(high)Foxp3(+) regulatory T cells via mechanisms that remain unknown. Here, we studied expansion, IL-2R-gamma chain signaling, survival pathways and resistance to apoptosis in human Treg responding to RAPA. METHODOLOGY/PRINCIPAL FINDINGS CD4(+)CD25(+) and CD4(+)CD25(neg) T cells were isolated from PBMC...
